Leonard (Johnny Galecki) y Sheldon (Jim Parsons) son dos físicos brillantes y compañeros de apartamento. Su vida se enreda cuando una preciosa joven, aunque poco inteligente, Penny (Kaley Cuoco, "8 Simple Rules", "Charmed"), se muda frente a ellos.

Para Sheldon, la llegada de Penny es un poco incómoda porque prefiere pasar sus noches jugando Klingon Boggle con sus amigos y compañeros de trabajo (también científicos) Wolowitz y Koothrappali.

Sin embargo, Leonard ve en Penny la posibilidad de aprender a interactuar con mujeres; siente que con ella se abre un nuevo mundo de posibilidades, y quizás hasta encuentre el amor.

Sheldon cree que este es un sueño que nunca alcanzará, pero quizás sus mentes brillantes puedan aprender algunas cosas de una joven que trabaja de mesera en "The Cheese Cake Factory".
