La hipótesis preferida es la de “La transferencia natural” que tiene como base a los chimpancés africanos, verdaderos portadores de esta enfermedad letal. Según este estudio, el virus se transfirió al hombre a través de una herida o mordisco, y luego se propagó entre los seres humanos. Otra teoría muy diferente establece la causa en el uso de una vacuna experimental de la polio, contaminada por el virus, y administrada a un millón de africanos en las colonias belgas, donde diez años después se encontró el primer caso de esta enfermedad.
